Karakterisasi Bakteri Penghasil Gas Metana pada Rumput Laut Jenis Eucheuma cottonii

Abstract


This study aims to determine the pressure of methane gas produced by the seaweed Eucheuma cottonii and characterization of methane-producing bacteria learn in the process of making biogas from Eucheuma cottonii seaweed species. Laboratory scale research conducted in July-August 2010. The material used is seaweed Eucheuma cottonii 5 kg and 10 kg of sediment taken from the coastal waters Kalianda, Lampung province. Making biogas and methane gas pressure calculations performed at the Laboratory of Marine Biology Department of Marine Science Sriwijaya University and characterization of bacteria carried in the Great Hall of the Public Health Laboratory Palembang. Measurement of methane gas pressure using a manometer and characterization of bacteria through the three stages of colony morphology, cell morphology and physiology with test observations of biochemical reactions. The results showed that the seaweed Eucheuma cottonii can produce methane gas with a maximum pressure of 14.90 psi on day 24. From the results obtained by characterization of the bacteria bacillus-shaped bacteria that are gram-negative group. In tests of biochemical reactions that function to see the metabolic activity of bacteria showed positive results in the motility test, glucose, maltose, indole, TSIA (H2S), red methil, citrat simmon’s, lysine decar and shown negative results in tests of lactose, mannitol, sucrose, urea, voges proskauers and ornithine decar.

ABSTRAK

Penelitian ini bertujuan untuk mengetahui tekanan gas metana yang dihasilkan rumput laut jenis Eucheuma cottonii dan mengetahui karakterisasi bakteri penghasil gas metana dalam proses pembuatan biogas dari rumput laut jenis Eucheuma cottonii. Penelitian skala laboratorium ini dilaksanakan pada bulan Juli-Agustus 2010. Bahan yang digunakan adalah rumput laut Eucheuma cottonii 5 kg dan sedimen pantai 10 kg yang diambil dari perairan Kalianda, provinsi Lampung. Pembuatan biogas dan perhitungan tekanan gas metana dilakukan di Laboratorium Biologi Laut Program Studi Ilmu Kelautan Universitas Sriwijaya serta karakterisasi bakteri dilakukan di Balai Besar Laboratorium Kesehatan Palembang. Pengukuran tekanan gas metana dengan menggunakan alat manometer dan karakterisasi bakteri melalui 3 tahap yaitu morfologi koloni, morfologi sel dan pengamatan fisiologi dengan uji reaksi biokimia. Hasil penelitian menunjukkan bahwa rumput laut Eucheuma cottonii dapat menghasilkan gas metana dengan tekanan maksimal 14,90 psi pada hari ke 24. Dari hasil karakterisasi bakteri didapatkan bakteri berbentuk basil yang merupakan kelompok dari gram negatif. Pada uji reaksi biokimia yang berfungsi untuk melihat aktivitas metabolisme bakteri menunjukkan hasil positif pada uji motilitas, glukosa, maltosa, indol, TSIA (H2S), methil red, simmon’s citrat, lysine decar dan hasil negatif ditunjukkan pada uji laktosa, manitol, sukrosa, urea, voges proskauers serta ornithine decar.
